Reactive astrocytes as imaged with 11C‐DED in patients with different dementia disorders

Background Neuroinflammation, which receives ever increasing interest in neurodegenerative diseases, comprises a heterogeneous cascade of events that are thought to be related the downstream neurodegeneration. The aim this study was evaluate binding 11C-Deuterium-L-Deprenyl PET (DED) as measure reactive astrocytes patients with different dementia disorders, and assess its association other disease biomarkers. Method Eleven clinical diagnosis semantic variant primary progressive aphasia (svPPA, n = 5) behavioral frontotemporal (bvFTD, 6) were recruited. All had cerebrospinal fluid biomarker profile inconsistent Alzheimer’s (AD) or negative amyloid-β scan. imaging protocol included 11C-DED-PET, 18F-FDG-PET, 3D T1 MRI. A group amyloid-beta positive AD (n 20) underwent similar investigations used for comparison. Z-scores created 11C-DED (Patlak slopes) tracer relative delivery (R1 parameter) healthy controls (HC; 20), assessing load cerebral perfusion, respectively. Result svPPA bvFTD showed significantly higher areas, compared HC. Patients temporo-occipital regional distribution consistent expected underlying pattern neurodegeneration those although across same diagnosis. Conclusion Reactive appear common feature reactivity differs. Ongoing work evaluates relationship between patterns astrocyte activation, glucose metabolism, atrophy, cognitive performance.
